dc.description.abstractEnThe flavescence dorée (FD) phytoplasma is a quarantine bacteria associated with a severe disease of grapevine and transmitted by the insect vector Scaphoideus titanus. In experimental conditions, FD phytoplasma is also transmitted by Euscelidius variegatus to broad bean in which it multiplies and also causes symptoms. To be transmitted to plants, phytoplasmas must invade different cell types of their insect vectors including midgut epithelial and salivary gland cells. The process of cellular endocytosis involves both bacterial and eucaryotic factors such as adhesins and receptors. FD phytoplasma expresses variable membrane (Vmp) proteins at its surface and the insect protein Uk1_LRR is one of the putative receptors of the adhesin VmpA (Arricau-Bouvery et al., 2021; Canuto et al., 2023). Once attached to insect cells, FD phytoplasma enters into eucaryotic cells using clathrin-mediated endocytosis (Arricau-Bouvery et al., 2023). In the present study, we have shown that VmpA-coated fluorescent beads enter E. variegatus cultured cells in a Uk1_LRR and clathrin dependent manner. In vivo experiments show that dsRNA mediated silencing of clathrin gene significantly reduced the colonization of E. variegatus by FD phytoplasma and the transmission to the plant Vicia faba. These results confirm the implication of clathrin in insect cells’ invasion by FD phytoplasma during E. variegatus colonization. Interestingly, silencing uk1_LRR gene increased the colonization of E. variegatus by FD phytoplasma without effect on the transmission to Vicia faba. We therefore hypothesize that uk1_LRR could be implicated in both FD phytoplasma recognition and immune response of E. variegatus toward FD phytoplasma entry into insect cells.
